摘要
利用Swiss-Model建立了克雷伯氏杆菌中FNR蛋白完整三级结构。模型的可靠性经Ramachandran图进行验证,分析发现模建蛋白85.8%残基φ和ψ角落在最佳区域,12.8%的残基落入其他许可区,0.5%的残基落在勉强许可区,0.9%的残基落在不允许区。经Scanprosite服务器对FNR蛋白功能域和活性位点进行搜索发现,FNR主要包括环核苷酸结合域和螺旋-转角-螺旋DNA结合域两个区域。同时对FNR蛋白中[4Fe-4S]簇结构的合理位置进行了探讨。
The tertiary structure of FNR protein was built by Swiss-Model.Reliability of the model was validated by the Ramachandran plot,which showed that φ and ψ angles of residues in most favoured regions were 85.8%,residues in additional allowed regions were 12.8%,residues in generously allowed regions were 0.5% and residues in disallowed regions were 0.9%,respectively.The domain and active sites of FNR were searched by ScanProsite Server.It was shown that FNR contained cyclic nucleotide-binding domain and the helix-turn-helix DNA binding domain.The reasonable position of [4Fe-4S] cluster was discussed.
