核盘菌侵染拟南芥的过程及其芥子油苷的变化 被引量:5

摘要 研究了核盘菌侵染拟南芥哥伦比亚生态型(Col-0)过程中,拟南芥的发病症状、活性氧和芥子油苷组分和含量的变化,及其抗性相关基因和芥子油苷生物合成相关基因表达的变化。结果表明,在侵染过程中,拟南芥的受侵染部位出现黄色水渍状病斑,随发病时间的延长病斑逐渐扩大到整株,其相对含水量逐渐下降,相对电导率不断上升,活性氧(过氧化氢和超氧阴离子)大量累积,植物细胞大量死亡;4-甲基硫氧丁基芥子油苷、总脂肪类芥子油苷和总芥子油苷含量逐渐下降,吲哚-3-甲基芥子油苷、4-甲氧基-吲哚-3-甲基芥子油苷和总吲哚类芥子油苷含量先上升后下降;抗性相关基因PR1和PDF1.2,以及吲哚类芥子油苷生物合成相关基因的表达都大幅度增强;抗性的诱导存在一定的系统性。研究表明芥子油苷,特别是吲哚类芥子油苷很可能参与了拟南芥对核盘菌侵染的防卫反应。 The symptoms and changes in the levels of reactive oxygen species(ROS) and glucosinolates,were studied in the course of Sclerotinia sclerotiorum infection of Arabidopsis thaliana Columbia(Col-0) ecotype as well as the expression of resistance-related genes and glucosinolate biosynthesis-related genes.Results showed that during the infection,yellow lesions appeared from the infection sites,and gradually expanded to the entire plant,causing lethal damage.The relative water content decreased gradually,while the relative electric conductivity increased,and ROS(hydrogen peroxide and superoxide anion) accumulated steadily,leading to plant cell death.The contents of glucoraphanin,total aliphatic glucosinolates,and total glucosinolates declined,but the contents of glucobrassicin,4-methoxyglucobrassicin and total indole glucosinolates increased at the beginning and then decreased.The expression of resistance-related genes and indole glucosinolates-related genes were greatly enhanced.The induction of resistance was partially systemic.These results suggest that glucosinolates,especially indole glucosinolates might be involved in the defense of Arabidopsis against S.sclerotiorum.
