摘要 目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)中雌激素受体(ER)和热休克蛋白90(HSP90)基因的表达情况。方法分离58例SLE患者和28例正常人新鲜外周血单个核细胞,提取总RNA,利用半定量RT-PCR方法逆转录并扩增ER和HSP90,通过凝胶图像扫描系统对PCR产物的电泳条带进行扫描分析。结果 SLE患者组PBMC中ER和HSP90mRNA表达明显高于正常对照组(P均<0.05),且活动期患者的表达高于稳定期患者(P均<0.05)。结论 SLE患者中存在ER和HSP90基因的过度表达,并与疾病活动性有关。 Objective To investigate the expression of estrogen receptor and heat shock protein 90 in peripheral blood mononuclear cell(PBMC)from SLE patients.Methods Fresh PBMC from 58 SLE patients and 28 healthy controls were isolated and total RNA were extracted.Then the mRNA expression of estrogen receptor and HSP90 were measured in semi quantitative with reverse transcriptase-Polymerase chain reaction(RT-PCR).PCR product bands were scanned and analyzed by gel electrophoresis system.Results The mRNA level of ER and HSP90 in the SLE patients were significantly higher than that in normal controls(P〈0.05).The mRNA level of ER and HSP90 in active SLE group were higher than that in inactive SLE group(P〈0.05).Conclusion ER and HSP90 in SLE patients is up-regulated,the overexpression of ER and HSP90 is associated with the disease activity
