摘要
目的确定植物凝集素亚慢性的效应谱,研究植物凝集素亚慢性中毒作用的靶器官,并提出安全限量参考值。方法通过阴道(雌鼠)或者直肠(雄鼠)注入含有赋形剂的植物凝集素。分低、中、高3个剂量组,每组12只小鼠,雌雄各半,剂量分别为0、1、2、4 mg/只,对照组给以相同剂量的润滑剂(避孕药赋形剂)。每日一次,共30d。所有动物均在第31天处死,测定各脏器系数。结果各组肝脏系数、脾脏系数和胸腺系数差异具有统计学意义。4 mg/只剂量组的肝脏系数为6.98±0.61,与对照组的5.32±0.31比较差异有统计学意义(P<0.01);2 mg/只剂量组和1 mg/只剂量组的肝脏系数分别为6.56±0.48和6.01±0.38,与对照组比较差异有统计学意义(P<0.05)。4 mg/只,2 mg/只及1 mg/只的脾脏系数分别为0.68±0.05,0.66±0.09及0.61±0.03与对照组的0.48±0.05相比差异有统计学意义(P<0.01)。4 mg/只的胸腺系数为0.29±0.02,与对照组的0.19±0.01比较差异有统计学意义(P<0.01),2 mg/只及1mg/只剂量组的胸腺系数分别为0.24±0.04及0.23±0.03与对照组的0.19±0.01比较差异有统计学意义(P<0.05)。其他脏器系数差异均无统计学意义。结论芸豆植物凝集素对小鼠阴道及直肠没有亚慢性毒性作用。
Objective To observe the subchronic toxicity of phytohemagglutinin(PHA) at different doses from Yunnan white kidney bean in mouse and provide the reference of dose limitation.Methods Different doses of PHA(at 4,2,1 mg/per mouse in high,middle,low dose groups and control respectively) in excipient were administered to vagina in 6 female and rectum in 6 male mice at 0.1-0.2 ml per mouse of each group every day.Same volume of excipient on control was administered by using the same way.Animals were killed after 30 days administration and their weight of main organs were recorded.Results There were significant differences in the indices of liver,spleen and thymus after the administration of PHA compared with those of the control.The liver index of 4 mg dose group was higher than that of the control(6.98±0.61 vs 5.32±0.31,P0.01).The liver indices of 2 mg and 1 mg dose groups were significant different(6.56±0.48 vs 5.32±0.31;6.01±0.38 vs 5.32±0.31,P0.05)as compared with those of the control.The spleen indices of all PHA dose groups were significantly different(P0.01)from those of the control(0.68±0.05,0.66±0.09 and 0.61±0.03 vs 0.48±0.05).The thymus index of 4 mg dose group was higher than that of the control(0.29±0.02 vs 0.19±0.01,P0.01).The thymus indices of 2 mg and 1 mg dose groups were significantly different(P0.01)as compared with those of the control(0.24±0.04 and 0.23±0.03 vs 0.19±0.01).There were no significant differences(P0.05) of other viscera indices after the administration of PHA when compared with those of the control.Conclusion That there is no subchronic toxicity of PHA from Yunnan white kidney bean in mouse by virginal and rectal administration.
