摘要
实验采用流式细胞术的检测分析方法,探究溴氯海因(BCDMH)对常见淡水藻类近头状伪蹄形藻的作用。研究结果显示BCDMH对近头状伪蹄形藻的生长抑制作用呈明显的浓度依赖型变化。根据不同浓度细胞生长抑制的百分率计算,24、48和72hBCDMH对近头状伪蹄形藻生长抑制作用的EC50分别为3.64、3.21和2.56mg·mL-1。生长抑制作用的无可观察效应浓度(NOEC)为0.31mg·mL-1。伴随着生长抑制作用,近头状伪蹄形藻细胞的活性氧(ROS)含量、酯酶活性及细胞膜完整性均发生了不同程度的改变,NOEC分别为0.16、0.31和1.3mg·mL-1。其中ROS含量的升高对BCDMH浓度的变化最为敏感,其次是酯酶活性的变化,细胞膜破损仅在5.0mg·mL-1和2.5mg·mL-1浓度组出现。结合本次研究结果,在日常使用BCDMH时,需要综合考虑使用环境和用途等因素,在不影响使用效果的前提下,应尽可能地降低其使用浓度。
Flow cytometry(FCM)was applied to analyze the effect of bromo-chloro-dimethylhydantoin(BCDMH)on Pseudokirchneriella subcapitata.The results showed a concentration-dependent growth inhibition effect of BCDMH on P.subcapitata.As the BCDMH concentration increased,the effect of inhibition profoundly rose.EC50 of BCDMH to P.subcapitata were 3.64 mg·L-1(24 h),3.21 mg·L-1(48 h),and 2.56 mg·L-1(72 h).No observed effect concentration(NOEC)of growth inhibition was 0.31 mg·L-1.Under the effect of growth inhibition,ROS content,esterase activity and membrane integrity varied;NOEC were 0.16 mg·L-1,0.31 mg·L-1 and 1.3 mg·L-1,respectively.Among all of the changes,increased ROS concentration was the most sensitive index to BCDMH concentration,followed by changes in esterase activity.A slight membrane damage only appeared in 5.0 mg L-1 and 2.5 mg·L-1 groups.Based on effect of application,minimizing the concentration should be recommended.
