金丝桃苷对小鼠胃粘膜损伤的保护作用及机制 被引量:21

The protective effect of hyperin on gastric mucosal injury in mice and its mechanism

摘要 目的研究金丝桃苷(Hyp)对小鼠乙醇性胃粘膜损伤的保护作用及其与氧自由基(OFR)、一氧化氮(NO)和前列腺素(PG)的关系。方法制备小鼠乙醇性胃粘膜损伤模型,测定胃粘膜损伤面积;测定血浆、胃组织内MDA、NO和PGE2含量。结果Hyp(20、40、80mg·kg-1,ip)可剂量依赖性地抑制无水乙醇诱发的小鼠胃粘膜损伤,其损伤抑制率分别为389%、496%和556%。Hyp可使小鼠胃粘膜损伤过程中血浆、胃组织中升高的MDA含量降低,使降低的NO水平回升。Hyp对血浆、胃组织中PGE2含量无明显影响。结论Hyp对小鼠乙醇性胃粘膜损伤具有保护作用,其作用机制与Hyp抗氧化作用和促进NO水平恢复正常有关。 Objective To study the protective effect of hyperin (Hyp) on gastric mucosal injury in mice induced by absolute ethanol (Eth) and relationship of the effect with OFR, NO and PG. Methods Make the model of gastric mucosal injury in mice induced by Eth (ig); the area of gastric mucosal injury in mice was determined; the contents of MDA, NO and PGE 2 of plasma and gastric tissue were measured. Results The area of gastric mucosal injury in mice was inhibited after administration of Hyp (20, 40, 80 mg·kg -1 , ip), the inhibitory rates were 38 9%, 49 6% and 55 6% respectively, The increase of MDA formation induced by Eth was significantly decreased by Hyp (20, 40, 80 mg·kg -1 ,ip), the decrease of NO content of plasma and gastric mucosal tissue was elevated to normal level, but Hyp did not obviously influence the PGE 2 contents of plasma and gastric tissue in mice. Conclusion Hyp could possess protective effect on gastric mucosal injury in mice induced by Eth. Its mechanism may be related to antioxidative effect and improvement of NO level to normal.
