摘要
核酶是一类具有催化活性的小RNA分子,可以特异性地识别并结合目标基因mRNA,使该mRNA断裂失活,从而阻断或降低目标基因的表达,起到基因沉默的作用。此外,核酶不会对细胞基因组产生任何副作用,因此核酶作为一种既安全又有效的分子生物学工具受到越来越多研究者的关注。核酶的这一特性可用于基因治疗,针对某些病原或肿瘤的基因设计特异性核酶,并将其导入细胞以阻断或降低这些基因在细胞内的表达,最终达到抑制病原增殖、肿瘤扩散的目的。作者就近几年核酶在人类和动物疾病治疗中的应用研究进展作一综述。
Ribozymes are catalytically active small RNA molecules,which can specifically recognize and bind mRNA of the target gene,and make it fractured and inactivated,thereby blocking or reducing the expression of target genes,and play a role as gene silencing.Besides,ribozymes do not have any adverse effect to cell genomes,so ribozymes as a safe and effective molecular biology tool are increasingly concerned.These properties of ribozymes can be used for gene therapy.For certain pathogens or tumor genes,design specific ribozymes and import them into cells to block or reduce the expression of these genes in cells,ultimately inhibiting pathogen multiplication and tumor proliferation.This article has reviewed the study progress on the treatment of human and animal disease using ribozymes in recent years.
