摘要
目的探讨新生大鼠心肌细胞二维培养过程中心肌细胞的纯化及心肌细胞与成纤维细胞的鉴别,为改进心肌细胞二维培养及心肌细胞三维培养提供依据。方法用不同差速贴壁次数、含Brdu培养心肌细胞,免疫荧光标记后用激光共聚焦显微镜分别对24h的细胞进行分析鉴定。结果不同培养方法对心肌细胞的纯化有影响。结论差速贴壁次数的不同得到的心肌细胞纯度不一样,加入适当浓度的Brdu可抑制成纤维细胞的生长,获得纯度更高的心肌细胞。
Objective AIM To investigate the neonatal rat myocardial cells in the course of the purification of two-dimensional culture and the identification of myocardial cells and fibroblast-like cells,and to provide basis of myocardial two-dimensional cell culture and three-dimensional cell culture.Methods The 24-hour cells that were cultured by different number of differential velocity adherent and the Brdu medium were analysis to identify respectively by laser scanning confocal microscope after immunofluorescence labeling.Results Differential velocity adherent technique has an effect on the purification of myocardial cells.Conclusion The number of Differential velocity adherent gets different purity of myocardial cells.The appropriate concentration of Brdu inhibits fibroblast growth and access to higher purity of myocardial cells.
