摘要
研究桑天牛不同地理种群的遗传分化,为探讨桑天牛在不同区域暴发危害的规律提供理论依据。采用AFLP技术,对6个桑天牛地理种群的DNA进行扩增,5对引物组合得到扩增条带共241条,有多态性条带196条,平均多态性位点比率为81.33%,其中杭州种群具有13条特异条带。遗传距离分析结果:来自河北省的4个桑天牛地理种群间的平均遗传距离为0.270 3;来自浙江杭州的桑天牛种群与来自河南济源的桑天牛种群之间的遗传距离为0.397 2,二者与河北省的4个桑天牛地理种群间的平均遗传距离分别为0.418 2、0.333 4。聚类分析也显示,地理上相距越远,桑天牛种群间的遗传分化越大。
Understanding genetic differentiation of various geographic populations of Apriona germari could provide theoretical basis for exploring the regularity of Apriona germari outbreak and damage in different areas.DNA sequences from 6 geographic populations of A.germari were amplified by means of AFLP technology.241 bands were obtained from amplifications with 5 primer pairs,including 196 polymorphic bands.The average percentage of polymorphic sites was 81.33%,among which populations from Hangzhou(Zhejiang Province) had 13 specific bands.Analysis on genetic distance revealed that the average genetic distance of 4 geographic populations from Hebei Province was 0.270 3.The genetic distance between Apriona germari populations of Hangzhou and Jiyuan(Henan Province) was 0.397 2.Both of them had an average genetic distance of 0.418 2 and 0.333 4 with the 4 geographic populations from Hebei Province,respectively.Clustering analysis also indicated that the farther the geographic locations,the higher the degree of genetic differentiation.
