摘要
目的 探讨 p5 3蛋白和 ras癌基因蛋白在白血病儿童脑脊液 (CSF)中的表达水平及与白血病化疗的关系。方法 应用 Western斑点印迹法检测 5 4例儿童急性白血病 CSF中 p5 3蛋白和 ras癌基因蛋白表达水平 ,并计算 p5 3 /ras比值。结果 白血病儿童危象期 CSF中 p5 3蛋白明显高于非肿瘤对照水平 (P<0 .0 1)。缓解期 p5 3蛋白和 ras癌基因蛋白与危象期比有增高趋势 (P<0 .0 1) ,同时高于非肿瘤对照水平 (P<0 .0 1)。白血病儿童 CSF危象期 p5 3 /ras比值明显升高 (P<0 .0 1) ,缓解期比值下降接近非肿瘤对照组 (P>0 .0 5 )。 17例白血病儿童化疗前后双份 CSF中 p5 3 /ras比值动态观察发现 ,化疗前比值升高 ,化疗后比值下降 ,其临床表现及预后好 ,如化疗前后比值持续在高水平不下降 ,临床表现差 ,预后不佳 ,骨髓近期将复发。结论 检测急性白血病儿童 CSF中 p5 3蛋白和ras癌基因蛋白 ,计算 p5 3 /ras比值变化有助于指导化疗、评定疗效、监测预后。
Objective To determine the expression levels of p53 protein and ras oncogene protein and calculated p53/ras ratio in CSF from children patients with leukemia as well as its correlation with the chemotherapy of leukemia in CSF from children patients with leukemia.Methods The expression levels of p53 protein and ras oncogene protein of fifty four cases of children patients with leukemia were detected by Western Blot in the present study.Results The levels of p53 protein in the crisis stage of CSF from children patients with leukemia were significantly higher than those non tumor comparison group (P< 0.01).The levels of p53 protein and ras oncogene protein in the remission stage were significantly higher than those in the crisis stage (P<0.01) and than those of non tumor comparison grou (P<0.01).The ratio of p53/ras of CSF from children patients with leukemia in the crisis stage were significantly elevated(P<0.01) and which in the remission stage were decreased toward the level of non tumor comparison group (P>0.05).Before and after chemotherapy,dynamic developing from ratios of p53/ras of a double share CSF from seventeen cases of children patients with leukemia,ratios before chemotherapy were elevated and after chemotherapy ratios were decreased.Its clinical symptoms and prognosis were well.As ratios before and after chemotherapy were comtinuously higher,its clinical symptoms and prognosis were bad and bone marrow would recurred in near perios.Conclusion The detection of p53 protein,ras oncogene protein and calculated p53/ras ratio in CSF from children patients with leukemia is of great use for guiding the treatment,evaluating the curative effect and estimating the prognosis.
