摘要
目的:观察姜黄素对肝纤维化大鼠肝组织抑凋亡细胞(Bcl-2)、促凋亡细胞(Bax)的影响,探讨其抗肝纤维化的机理。方法:采用四氯化碳皮下注射加乙醇灌胃制备大鼠肝纤维化模型,用免疫组织化学方法(SABC)检测各组大鼠肝组织中Bcl-2、Bax的含量。结果:与模型组比较,姜黄素大、中剂量组肝组织纤维间隔中Bcl-2的表达明显下降(P<0.05),Bax的表达无明显改变(P>0.05),Bax/Bcl-2比值明显升高(P<0.05)。结论:姜黄素可有效升高纤维间隔中Bax/Bcl-2比值,并能抑制肝星状细胞(HSC)活化增殖,诱导活化的HSC凋亡,具有治疗肝纤维化的作用。在一定有效剂量范围内,其抗肝纤维化作用强度与剂量无明显依赖性。
Objective:Observe the effect of Curcumin on Bcl-2 and Bax of hepatic fibrosis tissues in rats,to further discuss the mechanism of antifibrotic action.Methods:Rat models of hepatic fibrosis were established by hypodermic injection of Carbon tetrachloride and filling the stomach with alcohol.The content of Bcl-2 and Bax in liver tissue of rats were detected by immunohistochemical technique(SABC).Result:Compared with model group,the Bcl-2 expression drops obviously in the liver fiber gap of middle and big dosage group of Curcumin(P0.05),the Bax expression changes indistinctively(P0.05),the ratio of Bax/ Bcl-2 in fiber gap was decreased by large(P0.05).Conclusions:Curcumin can decreased the ratio of Bax/ Bcl-2 in fiber gap,then suppressed the HSC activation and multiply,and inducted activated HSC to apoptosis.The Curcumin has the function of treatment hepatic fibrosis,the relevance didn't present between anti-hepatic fibrosis function and the dosage.
