沙眼衣原体蛋白激酶CT145的克隆及表达

摘要 目的构建含沙眼衣原体(chlamydia trachomatis,Ct)基因CT145的真核重组表达质粒pcDNA4/CT703,并检测其在HeLa细胞中的表达。方法利用PCR技术扩增全长CT145基因,并将其亚克隆到真核表达载体pcDNA4,双酶切和测序检测重组质粒正确性。将正确的重组质粒转染HeLa细胞,并用免疫荧光和WesternBlot检测重组蛋白表达情况。结果酶切和测序结果证实成功构建了重组质粒pcDNA4/CT145,同时免疫荧光和Western Blot实验证实重组质粒转染HeLa细胞后能表达相应的蛋白。结论重组质粒pcDNA4/CT145的成功构建和在HeLa细胞中成功表达为进一步研究沙眼衣原体与宿主细胞的相互作用奠定了基础。
