摘要
根据GenBank公布的鸡β-防御素序列设计一对特异性引物,运用RT-PCR技术从新郑三黄鸡肺脏组织中扩增其β-防御素基因并测序分析,结果表明,获得了195bp的cDNA目的片段,编码65个氨基酸;BLAST分析表明,该片段与GenBank中已公布的鸡β-防御素gal-9序列基因同源性达到98%。其与已公布的10个gal-9序列进行同源性比较,并绘制系统发育进化树的结果显示,所测的防御素基因核苷酸同源性为91.8%~94.4%;进化树分析表明获得的目的基因与gal-9基因处于同一进化分支。这为进一步研究gal-9基因在地方品种鸡中的先天性免疫功能奠定了基础,也为研发具有广谱抗菌活性的新型肽类生物制品奠定了前期基础。
According to chicken β-defensin sequences published in GeneBank,a pair of specific primers was designed.The Gal-9 gene was amplified by using RT-PCR from the lung tissue of Xinzheng Sanhuang chicken.Then it was sequenced.The results showed that the length of the cDNA fragment obtained was 195 bp,encoding 65 amino acids;BLAST analysis showed that the homology between the fragment cloned and the chicken β-defensin- gal-9 gene sequences published in the Gene Bank was 98% .Then the sequence was contrasted with the 10 gal-9 sequences published,at the same time,the phylogenetic tree was drawn.The results showed that the homology of them is from 91.8% to 94.4%;Phylogenetic tree analysis showed that the gene and gal-9 gene in the same evolutionary branch.That not only provided a base for research on immunologic function of gal-9 in the native chicken,but also laid the foundation of developing new antimcrobial product with broad spectrum.
