摘要
目的研究水蛭素与阿霉素(ADM)联合应用对人舌鳞癌细胞株TCA8113生长的影响。方法通过MTT法测定其对TCA8113细胞的生长抑制作用;应用流式细胞术检测凋亡及分析细胞周期的变化;利用吖啶橙-溴化乙啶(Ao-EB)染色观察水蛭素作用前后细胞形态的改变;应用透射电镜观察药物对细胞超微结构的影响。结果水蛭素、ADM对TCA 8113有明显的抑制增殖和诱导凋亡作用,并呈时间-剂量效应(P<0.05);水蛭素(15ATU/mL)与ADM(0.39~3.125 mg/L)联合给药可对TCA 8113产生增强的联合杀伤效果(P<0.05);流式细胞术检测提示水蛭素可促使细胞凋亡并阻滞细胞周期于G0/G1期;透射电镜和荧光染色观察可见凋亡细胞染色质凝聚、边集,并出现凋亡小体。结论水蛭素和阿霉素均对TCA8113有抑制作用,水蛭素的作用机制可能为诱导细胞凋亡并阻滞细胞周期于G0/G1期。水蛭素可增强TCA8113对ADM的敏感性,对于阿霉素化疗有增效减毒作用。
Objective It is to observe the influence of hirudin combined with adriamycin(ADM) on anti-proliferation of tongue squamous cell carcinoma line TCA8113. Methods The inhibitory effect on cell proliferation was assessed by MTT assay.The cell cycle of TCA8113 cells was detected with Flow Cytometry(FCM).The change of cytomorphology of TCA8113 cells was observed by Ao-EB coloretur;the cell ultrastructure was observed by transmission electronmicroscopy. Results Hirudin and ADM could inhibit TCA8113 cell proliferation and induce apoptosis in a time-dose dependent manner(P〈0.05).The combination of Hirudin(15 ATU/mL) and ADM(0.39 mg/L to 3.125 mg/L) showed enhanced joint killing effect(P〈0.05).The blocking of the cell cycle was in G0/G1-phase.Apoptotic feature was observed by transmission electronmicroscopy and fluorescent staining. Conclusion Hirudin can significantly inhibit the proliferation of TCA8113 cells.The mechanism may be induce the apoptosis and prohibit the distribution of cell cycle.Hirudin can sensitize TCA8113 cells to adriamycin-mediated apoptosis.
