低温冷冻对视网膜色素上皮细胞形态及超微结构的影响

摘要 目的:明确冷冻对视网膜色素上皮细胞( R P E)形态及超微结构的影响。方法:用处于融合状态的原代及第三代 R P E细胞,放入- 70℃深低温冰箱内,分别冷冻15,30,45,60,120 s,应用相差显微镜、扫描及透射电镜观察细胞形态及超微结构的变化。结果:相差显微镜观察:冷冻使细胞形态改变,细胞变圆皱缩,细胞间缝隙加宽;扫描及透射电镜观察:冷冻使细胞连接断离,细胞膜双层结构改变,部分细胞的细胞膜破裂。结论:冷冻可导致 R P E细胞间连接断离,且与冷冻时间有关,冷冻超过60 s 使细胞破裂。 Objective:This paper was to observe the effects of freeze on morphological and ultrastructural changes in human retinal pigmental epithelial (RPE) cells. Methods:We froze cultured human RPE cells in vitro under -70℃ for 15,30,45,60,120 seconds in refrigerator. Phase contrast microscopy as well as scanning and transmission electron microscopy were used to observe the effects. Results:Under phase contrast microscopy, the cells were round and shrinking, and intercellular space widened. Under scanning and transmission electron microscopy, cells were separated, and cell membrane altered in its dual layer structure or partially broke. Conclusion:Freeze separated the intercellular connection in RPE cells. Cells broke in ultra cold over 60 seconds. Conclusion:Freeze separated the intercellular connection in RPE cells.
