不同侵袭潜能的骨肉瘤细胞中抑癌基因IDH1的表达 被引量:1

摘要 目的 观察抑癌基因IDH1在不同侵袭能力的人骨肉瘤MG-63、U20S细胞株中的差异表达。方法培养人骨肉瘤细胞MG-63、U20S,以实时定量PCR、免疫细胞化学染色法检测MG-63、U20S细胞中IDH1基因mRNA及蛋白的表达情况;体外侵袭实验测定两骨肉瘤细胞株的侵袭能力。结果IDH1基因mRNA及蛋白在不同侵袭能力的骨肉瘤细胞株中存在差异表达,其在低侵袭力细胞株U20S中的表达量明显高于高侵袭力细胞株MG-63中的表达量(P〈0.01)。结论IDH1的表达与骨肉瘤细胞的侵袭能力呈负相关,其可作为判断骨肉瘤侵袭能力有价值的指标。 Objective To investigate the IDH1 expression in osteosarcoma (OS) cell lines with different invasion potential and analyze its significance. Methods Expressions of IDH1 in Mg63 and U2OS cell lines were detected by immunocytochemistry and Reahime PCR. The invasion ability of OS cell lines was measured using transwell chamber with matrigle. Results Expressions of IDH1 mRNA and protein were lower in MG63 compared with U2OS. The penetrating cell number of MG63 were higher than U20S. Conclusion Expression of IDH1 is lower in MG63 with high invasion potential than in U2OS with low invasion potential. IDH1 may relate to invasion ability.
