摘要
RT-PCR检测发现,成人骨髓红系祖细胞在培养条件下胎儿珠蛋白基因高水平表达(BFUe集落细胞中γ/γ+β达43.2%~58.3%),比较培养条件与体内造血微环境的差异,提示细胞因子,特别是胎儿期特异的细胞因子可能是导致胎儿珠蛋白基因重新活化的原因。研究细胞因子对球蛋白基因开关的影响,可能提供一种应用细胞因子组合治疗β-地中海贫血和镰刀型贫血的方法。
Human adult bone marrow mononucear cells were cultured in EPO-contained IMDMmethyl cellulose semi-solid medium,then BFUe- and CFUe-derived cells (BFUeDCs and CFUeDCs)were collected. γmRNA and γ+β mRNA expressed in these cells were detected by RT-PCR. The γglobin genes were highly expressed in bone marrow BFUeDCs (γ/γ+β 43.2%-58.3%), while theirtranscripts were hard detected in normal human adult PBCs (γ/β+β<1%). Comparing with normalhuman adult PBCs, the high level of γ/γ+β implicates that the γ-globin gene may be reactivated bycytokines, especially by fetal stage-specific cytokines. This result indicates that fetal stage-specificcytokine combination may potentially be used to treat β-thalassemia and sickle cell anemia.
