摘要
目的 在皮下导入血管紧张素Ⅱ (AngⅡ )诱导的心肌重塑大鼠模型上评价AngⅡ 1型受体 (AT1)转录水平在心肌重塑中的作用。方法 将 18只 6周龄雄性SD大鼠随机分为三组 ,每组 6只 ,分别给予皮下埋藏装有生理盐水或AngⅡ的渗透微泵 ,分如下三组 :组 1(对照组 ) :输注生理盐水 ;组 2 :输注AngⅡ ,同时管饲DMP8113mg·kg-1·d-1;组 3 :输注AngⅡ ( 2 0 0ng·kg-1·min-1)。各组持续输注 1周后取其心脏 ,提取总RNA ,采用逆转录多聚酶链反应 (RT PCR)测量心肌AT1基因、胶原Ⅰ、Ⅲ基因的转录水平。在AngⅡ或生理盐水输注前和处死动物前分别测量尾动脉压 (BP)、心率 (HR)、体重 (BW )、以及处死后心重 (CW ) ,并计算心重 /体重比 (C/B)。结果 实验前后 ,三组间BP、HR、BW差异均无显著性。每组动物试验前后的BP、HR也无变化 ,AngⅡ输注组的CW、C/B、AT1、胶原Ⅰ、Ⅲ基因表达水平分别高于对照组 ( 4 7± 0 4 ) % ,( 4 9± 0 9) % ,( 2 4 7± 3 5 ) % ,( 2 2 0± 4 7) %和 ( 2 0 6± 4 9) % ,P均 <0 0 1,并且这些改变能被DMP811完全阻断。同时还发现 ,AT1mRNA水平与C/B及与Ⅰ、Ⅲ型胶原mRNA水平呈正相关 (P <0 0 1)。结论 用渗透微泵方法向大鼠皮下导入非加压剂量的AngⅡ 1周 。
Objective To study the role of the transcriptional level of angiotensin Ⅱ (Ang Ⅱ) receptor type1 (AT 1) mRNA in myocardial remodeling as well as the influence of DMP811 on myocardial remodeling in rats Methods Six week old male SD rats with osmotic minipump subcutaneously containing either Ang Ⅱ or saline were used The rats were studied in three groups: (1)Saline infused (control) group (n=6), (2) Ang Ⅱ infused and DMP811 treated (3mg·kg -1 ·d -1 , by gavage) group (n=6), (3)Ang Ⅱ infused group (200ng·kg -1 ·min -1 ) (n=6) Ang Ⅱ or saline was continuously infused for 1 week Systolic blood pressure of the three groups was measured by the tail cuff method DMP811 was given 24h prior and up to the end of AngⅡ infusion The mRNA levels of AT 1 and collagen subtype Ⅰ, Ⅲ were measured with RT PCR Results Blood pressure, heart rate and body weight were not significantly different among three groups before Ang Ⅱ infusion Cardiac weight, cardiac weight/body weight ratio (C/B), the mRNA levels of AT 1, collagen Ⅰ, Ⅲ were increased by (4 7±0 4)%, (4 9±0 9)%, (24 7±3 5)%, (22 0±4 7)% and (20 6±4 9)%, respectively ( P <0 01)in Ang Ⅱ infused group compared with those in control group The above changes caused by Ang Ⅱ were inhibited completely by DMP811 AT 1 mRNA level correlated positively to C/B and collagen Ⅰ, Ⅲ mRNA levels significantly ( P <0 01) Conclusions AngⅡ plays an important role in myocardial remodeling and the regulation of AT 1 mRNA level may be one of the important mechanisms via which Ang Ⅱ induces myocardial remodeling
