摘要
目的:探讨PIK3CA在胃癌组织中的表达及其与转移的关系。方法:采用实时定量PCR和免疫组织化学方法检测PIK3CA基因在正常胃黏膜组织、原发胃癌组织、淋巴结转移及远处转移胃癌组织中mRNA和蛋白表达水平,并比较各组间的差别及其与临床病理特征的关系。结果:淋巴结转移胃癌组织中PIK3CAmRNA表达水平最高,分别约为正常胃黏膜组织及原发胃癌组织的5倍和2倍,差异具有统计学意义(P<0.05),但与远处转移胃癌组织比较差异无统计学意义。免疫组化结果表明,PIK3CA蛋白在原发胃癌组织、淋巴结转移及远处转移胃癌组织中阳性率分别为35%、67%和61%。与原发胃癌组织比较,淋巴结转移及远处转移胃癌组织中PIK3CA蛋白表达差异具有统计学意义(35%vs67%,P=0.015;35%vs61%,P=0.044)。此外,胃癌组织中PIK3CA蛋白高表达与淋巴结转移有关(P<0.05),而与患者的性别、年龄、临床分期等因素无关(P>0.05)。结论:PIK3CA高表达可能在胃癌的发生发展中具有重要作用,检测PIK3CA表达水平有望成为评价胃癌转移的一个重要指标。
Objective To explore PIK3CA expressions in gastric cancer tissues and the correlation between PIK3CA expression and metastasis. Methods PIK3CA mRNA and protein levels of normal gastric mucosa, primary gastric cancer tissues, gastric cancer specimens in lymph node metastasis and distant metastasis were detected respectively by Real-time quantitative PCR and immunohistochemistry. In addition, the difference of PIK3CA expression in each group and their relationship with clinical pathological characteristics were analyzed. Results PIK3CA mRNA in lymph node metastasis group was the highest, which was approximately 5 and 2 fold, as that in normal gastric mucosa and primary gastric cancer tissues, respectively (P 〈 0.05). While was not statistically different with that in distant metastasis group (P 〈 0.05). The positive rate of PIK3CA protein expression was significantly higher in lymph node metastasis and distant metastasis gastric cancer specimens than that in primary gastric cancer tissues(67% vs 35%, P = 0.015; 61% vs 35%, P = 0.044). Additionally, high expression of PIK3CA protein in gastric cancer tissues was associated with lymph node metastasis (P 〈 0.05), but wasn't associated with other factors such as patient's sex, age, and clinical stage. Conclusion High expression of PIK3CA might play a vital role in tumorigenesis and progression of gastric cancer. Its expression level might be an important indicator to evaluate gastric cancer metastasis.
