摘要
目的考察大孔吸附树脂对柴胡总皂苷的吸附解吸性能,优选纯化柴胡总皂苷的最佳树脂;采用离子交换树脂对柴胡总皂苷进行进一步的脱色纯化,建立柴胡总皂苷的富集与纯化方法。方法采用静、动态吸附与解吸实验,优选出对柴胡皂苷吸附与解吸较好的NKA-9型大孔吸附树脂,用于除去水溶性杂质、富集总皂苷;采用弱碱性阴离子交换树脂D900对其进行进一步脱色纯化。通过高效液相色谱法和紫外分光光度法,测定富集与纯化各个阶段柴胡总皂苷含量的变化。结果 7种不同性质的大孔吸附树脂中,NKA-9树脂的吸附与解吸率相对较高。同时确定NKA-9树脂上样量和洗脱条件,洗脱率>95%。再经D900离子交换树脂脱色后,总皂苷纯度>70%。结论通过树脂联用技术可简便、快速地富集纯化柴胡总皂苷,为其工业化生产提供依据。
Objective To assess the absorbing and desorbing capability of macroporus resins for total saponins in Bupleurum chinense and optimize the macroporous resin for better purifying total saponins.To establish a method for enriching and purifying the total saponins from Bupleurum chinense by using an anion exchange resin to decolor the enriched total saponins. Methods Static and dynamic adsorption and desorption experiments were carried out to screen the optimal macroporous resin.NKA-9 resin was chosen to remove the soluble impurities and enrich total saponins.The weakly basic anion exchange resin D900 was used to give a further purification.The spectrophotometric assay and HPLC were used to detect the changes of total saponins. Results NKA-9 showed a better adsorption and desorption property among the seven types of macroreticular resin.The loading volume and eluting conditions were fixed simultaneously,and the elution ratio was over 95%.After being decolorized via anion exchange resin D900,the purity of total saponins reached to 70%. Conclusion The combination of resin can simply and fastly enrich and purify total saponins from Bupleurum chinense,which providing the basis for industrial manufacturing.
