补肾柔肝活血法对缺血性中风大鼠脑组织中NO、NOS的影响 被引量:6

摘要 目的:探讨补肾柔肝活血法对缺血性中风大鼠脑组织中NO、NOS的影响。方法:采用改良的Longa法建立大鼠左侧大脑中动脉梗阻(MCAO)再灌注模型,造模成功后随机分为6组,假手术组、模型组、西药组、中药高剂量组、中药中剂量组、中药低剂量组。给予相应药物治疗14 d后测定大鼠脑组织中一氧化氮(NO)及一氧化氮合酶(TNOS与iNOS)水平。结果:①与假手术组比较:模型组大鼠脑组织中NO,TNOS及iNOS的含量明显高于假手术组(P<0.05);②与模型组比较:各治疗组的NO、TNOS、iNOS的水平均有所下降,治疗各组脑组织内NO和iNOS水平均极明显低于模型组,有极明显的差异性(P<0.01),但中药低剂量组及中药高剂量组脑组织内TNOS水平与模型组无明显差异(P>0.05),中药中剂量组和西药组脑组织内的TNOS水平明显低于模型组,有明显差异性(P<0.05);③与西药组比较:中药各治疗组脑组织内的NOS水平与西药组无明显差异性(P>0.05),中药中剂量组脑组织内的NO水平均明显低于西药组,有明显的差异性(P<0.05),但中药高剂量组和中药低剂量组脑组织内的NO水平与西药组比较,差异无显著性(P>0.05)。结论:补肾柔肝活血法治疗能明显降低大鼠脑组织中NO、NOS水平。 Objective: To evaluate the effect of Bushen Rougan Huoxue Method on NO,NOS in brain tissue of rat with ischemic stroke.Methods: The modified Longa technique was used to make reperfusion model rats with middle cerebral artery obstruction(MCAO) in the left brain,then the models were randomized into 6 groups: sham operation group,model control group,comparison group of Western medicine,high-dose group of Chinese drugs,middle-dose of Chinese drugs,low-dose group of Chinese drugs.Nitric oxide(NO) and nitric oxide synthase(TNOS and iNOS) levels in rat brain tissues were measured 14 days after the corresponding drug administration.Results: ① The content of NO,TNOS and iNOS of rat brain tissues in the model group was significantly higher than that of the sham operation group(P0.05);② The levels of NO,TNOS,iNOS decreased in treatment groups,and their levels were significantly lower than that of the model group,there was a very significant difference(P0.01),but there was no significant differences(P0.05) in TNOS levels of brain tissues in the low-dose group and high-dose group compared with the model group.TNOS levels of brain tissues in the middle-dose group of Chinese drugs and Western medicine were significantly lower than those of the model group,there were significant differences(P0.05).③ There was no significant difference between treatment groups of Chinese drugs and Western medicine group(P0.05) in TNOS levels of brain tissues.there was significant differences(P0.05) in TNOS levels of brain tissues in the middle-dose group of Chinese drugs and Western medicine group,but there was no significant differences(P0.05) in TNOS levels of brain tissues in the low-dose group and high-dose group compared with Western medicine group.Conclusion: Bushen Rougan Huoxue Method can significantly reduce NO,NOS levels in brain tissues.
