摘要
目的研究肝细胞癌(HCC)患者外周血CD4+CD25+调节性T细胞(Treg)和肝癌组织叉状头/翅膀状螺旋转录因子(FOXP3)的表达,并分析其与肝细胞癌临床病理的关系。方法采用流式细胞仪对HCC患者和健康志愿者外周血CD4+CD25+Treg细胞进行检测,免疫组化、Western blotting法检测FOXP3蛋白表达水平,并结合HCC患者临床病理特征进行分析。结果 HCC患者外周血CD4+CD25+Treg表达率为(16.76±5.53)%,较健康志愿者的(6.04±2.01)%显著增高(P<0.01)。免疫组化染色肿瘤组织FOXP3阳性细胞数为(103.95±34.31)个/mm2,高于癌旁组织的(29.61±4.65)个/mm2(P<0.05),相应肿瘤组织FOXP3蛋白表达也增高。CD4+CD25+Treg细胞水平与肿瘤多灶性有关。结论 HCC患者的外周血CD4+CD25+Treg细胞表达增高,FOXP3蛋白在肿瘤组织中表达与CD4+CD25+Treg细胞的变化相平行,可能与肿瘤多灶性有关。
Objective To investigate the levels of CD4+CD25+regulatory T cells(Treg) and forkhead/winged helix transcription factor(FOXP3) and their clinical significance in hepatocellular carcinoma(HCC) patients.Methods Peripheral blood mononuclear cells(PBMCs) from HCC patients and healthy controls were analyzed for Treg frequency using flow cytometry.The expression of FOXP3 protein in tumor tissue was examined by immunohistochemistry and Western blotting.Results The proportion of Treg cells in peripheral blood was significantly higher in HCC group than in control group[(16.76±5.53)% vs.(6.04±2.01)%,P〈0.01].The number of FOXP3 positive cells was higher in HCC group than in paracancerous group[(103.95±34.31)/mm2 vs.(29.61±4.65)/mm2,P〈0.05].The expression of FOXP3 protein also increased in HCC.The variation in the number of Tregs correlated positively with that of more foci in tumors.Conclusion The proportion of Treg cells in peripheral blood with HCC patients increased.The levels of FOXP3 protein also increased in HCC,which paralleled the variation in the number of Tregs.The variation in the number of Tregs correlated positively with that of more foci of tumor.
