摘要
目的 观察脂质体携载C型利钠利尿肽(CNP) 对大鼠主动脉血管舒张、平均动脉血压和内皮素(ET) 刺激主动脉平滑肌细胞增殖的影响。方法 主动脉环灌流测定血管张力;平滑肌细胞培养;[3H]TdR参入测定DNA合成;反相蒸发法制备脂质体。结果 脂质体携载CNP的舒张主动脉和降低平均动脉压作用显著强于单用CNP。ET刺激平滑肌细胞增殖,使VSMC[3H]TdR 参入量增加1-8 倍。10 -8 、10- 7 、10- 6 mol·L-1 CNP可使ET 刺激[3H]TdR参入量分别下降2% ( P> 0-05), 21 % ( P< 0-05) 和55% ( P<0-01) ,而同样浓度的脂质体携载CNP 使ET 刺激VSMC[3H]TdR参入量分别下降56% ( P< 0-01),59% ( P<0-01) 和66 % ( P< 0-01), 其作用强于CNP。脂质体和CNP对[3H]TdR 参入量与对照组相比差异无显著性。结论 脂质体携载CNP的舒张血管、降低平均动脉压和抑制ET
AIM In this study, we observed the effect of liposome encapsulated C type natriuretic peptide (CNP) on aortic relaxation, mean arterial blood pressure and endothelin (ET) induced DNA synthesis in cultured vascular smooth muscle cells (VSMCs) of rat. METHODS Perfusion of arterial rings; rat VSMC culture; DNA synthesis was assayed by [ 3H] TdR incorporation; Liposomes were prepared by reverse evaporation from a mixture of egg yolk phosphadyl choline, cholesterol and DOPC (molar rato7∶2∶1). RESULTS Compared with unencapsulated CNP, encapsulated CNP in liposome relaxed rat aortic rings and decreased mean arterial blood pressure greater. ET enhanced the cultured VSMCs [ 3H] TdR incorporation by 1 8 fold. 10 -8 ,10 -7 ,10 -6 mol·L -1 CNP decreased VSMC [ 3H] TdR incorporation induced by ET 2% ( P >0 05), 21% ( P <0 05) and 55% ( P <0 01), but equivalent concentrations of the encapsulated CNP in liposome significantly reduced VSMC [ 3H] TdR incorporation by 56% ( P< 0 01), 59% ( P <0 01) and 66% ( P <0 01), and unencapsulated CNP and liposomes were without significant effect on the VSMCs [ 3H] TdR incorporation. CONCLUSION The magnitude of the encapsulated CNP in liposome relaxation of aortic rings, reduction of mean arterial blood pressure and inhibition of VSMC proliferation significantly greater compared with equivalent concentration of the unencapsulated peptide
