摘要
用国产抗肿瘤药物三尖杉酯碱处理HL60 和L12 10 细胞 ,探讨药物对两种不同的白血病细胞以及在不同条件下培养的同种白血病细胞核DNA结构的影响。DNA凝胶电泳结果表明 :(1) 0 2 μg/mlHT作用 2h的HL60 细胞和HT作用 2~ 2 4h的L12 10 细胞有明显的DNAladder。 (2 )HT处理 6~ 2 4h的HL60 细胞、在DBA/ 2纯种小鼠体内培养的被HT处理不同时间的L12 10 细胞及所有对照细胞无DNAladder。 (3)HT处理 2 4h的HL60 细胞和HT处理 12h的白血病小鼠体内的L12 10 细胞核DNA断裂成碎片 ,在琼脂糖凝胶上呈弥散状分布。
The authors reported the nuclear DNA structure changes of the leukemic HL 60 cells and L 1210 cells induced by antitumor drug harringtonine(HT) at 0.2μg/ml and 20μg/mouse with different induction peridos (0 h,2 h,6 h,12 h,24 h).The results of agarose gel electrophoresis of DNA showed the “DNA ladder”in HL 60 cells induced with HT for 2 h and in L 1210 cells induced with HT for 2~24 h, with no DNA ladder in HL 60 cells induced with HT for 6~24 h and in L 1210 cells induced with HT for 2~24 h in DBA/2 mice and in all the control, with DNA fragmentations in the HL 60 cells induced with HT for 24 h and L 1210 cells induced with HT for 12 h in the DBA/2 mice.
