摘要
目的观察孕鼠被动吸烟对胎鼠海马神经元一氧化氮合酶表达的影响,以期为探讨被动吸烟对子代危害的机制提供更多实验依据。方法将雌性大鼠随机分为对照组和实验组两组。实验组采用人工熏烟的方法建立被动吸烟模型,对照组不作处理。妊娠21日人工结束分娩,取胎鼠脑组织,进行一氧化氮合酶(NOS)免疫组化染色。高倍镜下对海马区观察并摄片,用图像分析软件Image-Pro Plus对两组切片进行光密度分析,观察其差别。结果 NOS在胎鼠海马区有表达,分布于海马神经元胞质内。计算机图像分析结果表明,与对照组比较,实验组海马NOS表达下降,统计结果有显著差异性(P<0.05)。结论孕鼠被动吸烟使胎鼠海马NOS的表达下降,其可能为孕期被动吸烟导致子代学习记忆记忆功能下降的机制之一。
Objective This study was designed to observe the effects of passive smoking on the expression of nitric oxide synthetase(NOS) of fetal rat hippocampal neurons,in order to get more laboratorial testification of the mechanism of the harm of passive smoking on filial generation.Methods Female rats were randomly divided into the test group(TG) and the control group(CG).TG was subjected to passive smoking,CG was not.The female rats were terminated pregnancy at the 21st day of pregnancy.The expressioin of NOS of fetal rat hippocampal neurons was observed and analysed by immunehistochemical technology.Results NOS was expressed in cytoplasm of fetal rat hippocampal neurons.The result of computer image analyse show that the expression of NOS of TC fetal rat hippocampal neurons descended,and there was significant statistical difference between TG and CG(P 0.05).Conclusion The expressioin of NOS of fetal rat hippocampal neurons descended as a result of passive smoking on pregnant rats,which may be one of the mechanism that passive smoking affect study and memory abilities of filial generation.
