实时荧光定量PCR检测宫颈癌组织中HGF mRNA和c-Met mRNA的表达 被引量:2

摘要 目的检测宫颈癌组织中肝细胞生长因子(HGF)及其受体原癌基因c-Met的分子表达水平,并探讨其临床意义。方法采用实时荧光定量PCR(QRT-PCR)技术检测43例宫颈癌,30例宫颈上皮内瘤变Ⅲ和27例正常宫颈组织标本中HGF和c-Met的分子表达水平。结果 HGF mRNA和c-Met mRNA在宫颈癌组织中均呈高表达,其与宫颈癌患者临床分期、肿瘤直径大小、浸润深度、淋巴结转移有关(P均<0.05),而与肿瘤的分化程度无关(P>0.05)。HGF mRNA和c-Met mRNA之间存在正相关(r=0.666,P<0.01)。结论 QRT-PCR技术可对宫颈癌组织中的HGF mRNA和c-Met mRNA表达进行定量检测;HGF mRNA和c-Met mRNA的高表达与宫颈癌的侵袭转移有关。两者联合监测有利于早期诊断宫颈癌,为宫颈癌的治疗和预后提供参考。 Objective To investigate the expressions of hepatocyte growth factor(HGF) and its receptor proto-oncogene c-Met in cervical carcinoma and their clinical significance.Methods The expressions of HGF mRNA and c-Met mRNA were detected by real-time quantitative reverse transcription-polymerase chain reaction(QRT-PCR) method in 43 cervical carcinoma tissues,30 cervical intraepithelial neoplasm(CIN) Ⅲ tissues and 27 normal cervical tissues.Results The positive expressions of HGF mRNA and c-Met mRNA increased remarkably in cervical carcinoma tissues(P0.05).In cervical carcinoma,the expressions of HGF mRNA and c-Met mRNA were correlated with the clinical stage,tumor size,invasion of deep muscular layer and lymph node metastasis(P0.05),but not with the grade of tumor pathology(P0.05).There was a significant positive correlation between HGF and c-Met in cervical carcinoma(r=0.666,P0.01).Conclusion QRT-PCR used to detect the expressions of HGF mRNA and c-Met mRNA may be an accurate and reliable means for early diagnosis of cervical carcinoma.Over-expression of HGF and c-Met mRNA may both be involved in the invasion and metastasis of cervical carcinoma.Monitoring HGF mRNA and c-Met mRNA would facilitate the treatment and prognosis of cervical carcinoma.
