摘要
直接将细胞色素C吸附在硝酸纤维素(NC)膜上,进行了基质辅助激光解吸电离- 飞行时间- 质谱(MALDI-TOF-MS)分析。质量准确度和重现性没有明显的降低,检测极限可达lpm ol。扫描电镜结果表明,本文所用NC膜具有孔径均匀、孔深适宜的特点,允许蛋白质和基质在膜上形成良好的“包裹型”或“包围型”
Cytochrome C absorbed on nitrocellulose(NC) membrane was directly analyzed by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI TOF MS).Mass accuracy and reproducibility are the same as those of standard sample preparation with the detection limitation of l pmol.Scanning electron microscopy showed that NC membrane enabled formation of protein matrix co crystalline because of its low mean pore sizes and not too deep pores.
