摘要
目的建立凝胶色谱法测定头孢地尼中聚合物的方法。方法色谱柱为Sephadex G-10柱(15.0mm×300mm),流动相A为pH7.0的0.1mol/L磷酸盐缓冲液,流动相B为水。流速为2.0mL/min,检测波长为254nm,进样量为200uL。结果该方法在4.1~20.3mg/mL范围内与峰面积呈良好的线性关系(r=0.9999),头孢地尼中聚合物含量小于0.1%。结论该方法简便、准确、灵敏度高,适用于头孢地尼聚合物的控制。
Objective To establish a gel filtration chromatography method for determination of the polymer in cefdinir. Methods The chromatographic column was Sephadex G-10 (15.0mm×300mm); mobile phase A, 0.1mol/L phosphate buffer (pH7.0); B, water; the flow rate was 2.0mL/min; the detection wavelength was 254nm and the injection volume was 200uL. Results The good linearity was obtained between peak area and concentration ranging from 4.1-20.3mg/mL, the content of polymer in cefdinir is less than 0.1%. Conclusion The method is simple, accurate and sensitive for the control of the polymer in cefdinir.
