摘要
应用TPO依赖细胞株检测rHu TPO体外生物学活性。将TPO标准品和样品分别稀释到一定的浓度 ,加入到 96孔细胞培养板中 ,每孔加入TPO依赖细胞 ,在 37℃CO2 孵箱中培养 48h后 ,加入一种四唑化合物 ,再培养6± 2h ,在 492nm波长下测A值。用TPO标准品做对照。根据剂量反应平行线法 ,计算出样品相对的体外生物学活性。该方法重复性好 ,回收率高 。
A method for detecting the in vitro biological activity of rHu TPO with TPO dependent cell strains was developed. The standards and samples of TPO were diluted to certain concentrations and added to a 96 well tissue culture plate. After TPO dependent cells were added in each well, the plate was cultured in CO 2 incubator at 37℃ for 48 h, and further cultured for 6±2 h after MTS was added in. The in vitro biological activities of samples were calculated out with A 492 values by dose respones paralled line method, using TPO standard as control. The method showed good reproducibility and high recovery rate, and was suitable for various laboratories.
