黄竹清脑颗粒对痰热腑实证脑出血大鼠急性期脑组织中Caspase-3表达的影响

摘要 目的:探讨黄竹清脑颗粒对痰热腑实证脑出血模型大鼠急性期脑组织中半胱氨酸蛋白酶(Caspase-3)表达的影响。方法:采用SD大鼠自体粪便灌胃配合胶原酶Ⅶ-肝素混合液立体定位注射的方法复制痰热腑实证脑出血大鼠模型,免疫组化法测定大鼠脑组织中Caspase-3表达的变化。结果:①黄竹清脑颗粒可明显减轻痰热腑实证脑出血大鼠喉中痰鸣、鼻分泌物多,粪便干结,烦躁等症状,并可改善大鼠脑出血后的神经体征;②黄竹清脑颗粒大、中剂量组明显优于清开灵颗粒组(P<0.05)。结论:黄竹清脑颗粒能降低大鼠脑出血后血肿周围神经元活化caspase-3的表达,阻止神经元的凋亡,可能是其保护脑神经细胞的作用机理之一。 Objective:To research the effect of the expression of Caspase-3 by HuangZhu QingNao granule,on experimental Intracerebral Hemorrhage with TanRe FuShi syndrome in rats.Methods:The method that the select SD ratadopt to type Ⅶ collagenase and heparin were injected into the right caudate nucleus of rats and auto-feces oral administration were used to develop the animal model of intracerebral hemorrhage with TanRe FuShi syndrome.The expression of caspase-3 in cerebral tissues was detected by immunohistochemistry technique.Results:①HuangZhu QingNaog ranule can significantly reduce symptoms as the phlegm chirps behave in the throat nose secretions,feces,anxiety,irritability in rat,and can improve the neural physical sign after cerebral hemorrhage;② large and medium HuangZhuQingNao granule group superior to clear to turn on fairy particle group(P0.05).Conclusion:during acute stage of intracerebral hemorrhage neural activation up-Caspase-3 obvious,HuangZhu QingNao granule can obviously decrease the expression of Caspase-3,Block apoptosis,neruonmay be a protective mechanism of brain cells.
