摘要
目的建立HPLC示差折光分析法测定乳糖的含量。方法采用Alltima-NH2柱,流动相为乙腈-水(67∶33),流速为1.0mL.min-1,柱温为45℃。结果乳糖在0.22~1.76mg.mL-1范围内线性关系良好(r=0.9991),平均回收率为99.0%,RSD为1.5%(n=9)。结论该法简便、快速,测定结果准确。
OBJECTIVE To establish a method for determination of Galactose content by HPLC with refractive index detector.METHODS Alltima-NH2 column was used.The mobile phase was acetonitrile-water(67∶33).the flow rate 1.0mL·min-1 and the column temperature 45℃.RESULTS The linear range for Galactose was 0.22~1.76mg·mL-1(r=0.9991),the average recovery was 99.0%(n=9),with RSD 1.5%.CONLUSION The method simple,timesaving and accurate.
