摘要
目的探讨转化生长因子-β(TGF-β)和甲状旁腺激素相关蛋白(PTHrP)对大鼠乳腺癌骨转移发生、发展的意义。方法建立大鼠乳腺癌骨移植模型,观察建模后影像学变化及TGF-β和PTHrP蛋白表达。结果随建模时间的延长,X线片显示胫骨骨皮质破坏加重,TGF-β和PTHrP蛋白在正常组织及建模后第7、14和21天的光密度值分别为912±246、1 586±483、4 254±850和7 026±1 033;946±256、1 932±393、3 128±809和4 653±1 008,两两比较,差异均有统计学意义(P<0.05)。结论 TGF-β和PTHrP可促进大鼠乳腺癌骨转移的发生、发展。
Objective To examine the expressions of the transforming growth factor-beta(TGF-β)and parathyroid hormone-related protein(PTHrP)on a rat model of metastatic bone breast cancer and to evaluate the significance.Methods The rat model of metastatic bone cancer was initiated by inoculations of the MRMT-I rats breast carcinoma cells in Sprague-Dawley rats.After establishing the model,segments of metastatic bone cancer were extracted to detect TGF-β protein and PTHrP protein expression through the immunohistochemical SP method and the bone structural damage,respectively,were monitored by radiological analysis.Results Seven,fourteen and twenty-one days after establishing the model,X-ray showed that tumor destroyed the bone more severely.The optical density value of the expressions of TGF-β and PTHrP protein were 912±246,1 586±483,4 254±850,7 026±1 033;946±256,1 932±393,3 128±809,4 653±1 008(P0.05)respectively in the normal tissue,7,14 and 2l days after establishing the model.Conclusion TGF-βand PTHrP can promote rat breast cancer occurrence and development of bone metastases.
