摘要
目的:研究丁香苦苷固体脂质纳米粒在鸭体内抗鸭乙肝病毒的作用。方法:采用实时荧光定量PCR方法筛选出先天感染DHBV阳性2日龄浙江麻鸭30只。将阳性鸭随机分成5组,分别是空白对照组、拉米夫定组、丁香苦苷固体脂质纳米粒低、中、高剂量组,每组6只,连续给药15天以后停药观察5天。在用药前、用药第51、0、15天及停药后第5天从腿胫静脉抽血,检测血清DHBV-DNA的含量,并检测血清的ALT、AST水平,停药5天后取肝脏组织进行病理学观察。结果:与空白对照组相比,丁香苦苷固体脂质纳米粒高、中剂量组的DHBV-DNA含量都明显下降,有显著性差异(P<0.05),ALT、AST水平也明显降低,并有减轻肝脏炎症反应的作用,作用的强弱与用药剂量相关。而丁香苦苷固体脂质纳米粒低剂量组未表现出明显的上述作用。结论:丁香苦苷固体脂质纳米粒在鸭体内具有抗鸭乙肝病毒的作用以及对DHBV引起的肝损伤具有保护作用。
Objective : To determine the therapeutic effect of SYR - SLN on DHBV in vivo and compare it with lamivudine in the treatment of DHBV so as to explore the possible mechanisms of this Chinese herb. Methods : Thirty model Zhejiang brown spot ducklings with congenitally infection of DHBV detected by PCR were divided into five groups randomly, which were black control group, high dose group, medium dose group and low dose group of solid lipid nanoparticles loaded with syringopicroside, as well as lamivudine group. They were treated with different doses of SYR -SLN and lamivudine respectively. At the day before the treatment, the 5th, 10th, 15th days in the treatment and 5th day after the suspend of it, the contents of serum DHBV - DNA were determined respectively by the means of quantitative real - time PCR. Serum levels of ALT, AST and liver biopsy were also done before and after the treatment with HE staining to detect the transformation of inflammation. Results: The levels of serum DHBV- DNA dropped markedly in high dose group and medium dose group of solid lipid nanoparticles loaded with syringopicroside (P 〈 0. 05, compared with black control group), the levels of ALT, AST were also dropped markedly, and had to reduce inflammation in the liver. Effects related to the dosage ; however there was no significance in low group. Conclusions : SYR - SLN does have therapeutic effect on DHBV hepatitis. It can not only restrain the duplication of DHBV in vivo but also eliminate inflammation in liver.
