摘要
通过建立的一种简单的反硝化聚磷菌筛选方法,获得一株源自池塘底泥的反硝化聚磷菌RC11,利用细菌自动分析鉴定仪等方法对其进行种属鉴定,确定其分类地位。试验结果表明,菌株RC11具有合成聚β-羟基丁酸和多聚磷酸盐的能力,硝酸盐还原阳性;在好氧情况下,可以直接利用培养基中的有机物(如乙酸、蛋白胨)氧化产能,吸收培养基中的磷酸盐合成多聚磷酸盐;能够利用硝酸盐进行反硝化聚磷,但在高浓度亚硝酸盐存在时,则表现为释磷;硝酸盐和乙酸钠同时存在于培养基中时,也出现缓慢的吸磷现象。经鉴定,菌株RC11属假单胞菌属细菌。
A simple method of isolation of denitrifying phosphate-accumulating bacterium from pond sediment was established in this paper,and a strain RC11was identified by autoanalyzer.The strain RC11 was showed to be capable of synthesis of PHB and poly-p of denitrification.The strain RC11 was found to be able to acquire energy from oxidizing external carbon resource using oxygen as electron accept directly to produce polyphosphate,and to acquire energy from oxidizing PHB using nitrate as an electron acceptor to produce polyphosphate.However,the strain RC11 resulted in releasing phosphate from cells at high concentration of nitrite.In the presence of nitrate and acetic acid,denitrifying and phosphate-accumulating were also observed,indicating that the strain RC11 was identified as Pseudomanas pp.
