摘要 目的建立动静力失衡性大鼠颈椎病模型以观察和探讨TNF-α、IL-1β在颈椎间盘退变组织中的表达和意义,进而探讨颈椎间盘退变的发生机制,为颈椎病的临床治疗提供实验依据。方法SD大鼠40只,被随机分为颈椎病动物模型组和假手术对照组,其中模型组20只,对照组20只。模型组大鼠切断浅层和深层颈肌并切除1.5 cm长一段,同时切断棘上、棘间韧带建立动静力失衡颈椎病动物模型。采用免疫组化方法检测模型组退变颈椎间盘和正常对照组颈椎间盘中的INL-α和IL-1β的表达,并进行组间数据的统计学分析。结果颈椎病模型组的颈椎间盘组织形态学较假手术对照组有明显的退行性变化。免疫组化法检测模型组颈椎间盘组织中的TNF-α及IL-1β的表达较假手术对照组显著增高(P<0.01)。结论模型组大鼠颈椎间盘较假手术对照组出现典型的退变形态学变化,细胞因子TNF-α和IL-1β在颈椎间盘中呈高表达可能与颈椎间盘退变相关。 Aim To set up the cervical disc rat model with unbalance of dynamic and static forces for observing and discussing the expression and significance of TNF-α,IL-1β in the cervical disc regression,moreover,for discussing the mechanism of the cervical disc regression in order to provide experimental evidence for the clinical treatment of cervical spondylosis.Methods Forty SD rats were randomly divided into 2 groups,20 for the animal model group with cervical spondylosis,and 20 for the comparison group with sham-operation.The model group should be cut off 1.5 cm superficial and deep layer neck muscle and cut off the supraspinatus and interspinal ligamentum for setting up the cervical disc rat model with unbalance of dynamic and static forces.The difference of expressing of TNF-α,IL-1β were compared between model group and normal comparison group with immunohistochemistry and analyzed.Results Compared with the sham-operation group,there was obvious regression change of intervertebral histomphology in the model group.The expression with immunohistochemistry of TNF-a and IL-1β in the model group was obviously higher than that in the normal group.There were statistical significances(P0.01).Conclusion Compared to sham-operated control group,model group rats cervical disc have typical degeneration morphology change,high expression of cytokine TNF-α and IL-1β in cervical disc may depend to cervical disc degeneration.
