摘要
目的采用大鼠切口疼痛模型,观察鞘内联合应用盐酸奈福泮和新斯的明的痛行为及对脊髓c—fos蛋白表达的影响。方法在大鼠切口疼痛模型上结合鞘内置管技术,采用累积疼痛评分法评定大鼠疼痛行为;运用免疫组织化学技术观察奈福泮、新斯的明及联合应用时对脊髓背角c—fos基因表达的影响。结果与F组相比,手术组大鼠累积疼痛评分均明显升高(P〈0.01);与s组相比,D,组、D:组及c,组均明显降低累积疼痛评分(P〈0.01);各组用药无统计学差异。奈福泮与新斯的明ED。。量单独使用时均明显抑制12-fos蛋白的表达,而1/2ED30两药的合用,抑制程度更大。它们的作用主要产生在脊髓Ⅰ~Ⅱ层、V~Ⅵ层,以Ⅰ~Ⅱ层分布最多。结论鞘内注射盐酸奈福泮和新斯的明的抗伤害作用与抑制c—fos蛋白的表达有关,在脊髓水平上,盐酸奈福泮与新斯的明对c—fos的抑制作用存在协同效应。
Objective To observe the effect of intrathecal nefopam and neostigmine on nociceptive behaviors and c-fos protein expression at spinal dorsal horn in a rat model of incision pain. Methods In a rat model of incision pain, cumulative pain scores were used to evaluate pain behaviors; Immunohistochemistry technique was used to observe c-fos protein expression at spinal dorsal horn. Results The cumulative pain scores in operation group in rats were much higher than those in sham -operation rats in intrathecal group. In groups D1, D2 and C1 the cumulative pain scores were decreased significantly after operation, but there were no statistical differences among the groups with drugs. When nefopam and neostigmine were used alone at the dose of ED50, both of them can significantly repress the expression of e-fos protein. When the two drugs were used together at the dose of 1/2 ED50, the repression degree was greater than that with nefopam or neostigmine. The acted positions were at the layer I-lI and V-VI in dorsal horn. Conclusion The antinociceptive effects of intrathecal nefopam and neostigmine are mediated partly by inhibition of c-fos protein, which reveals synergistic interaction of neotigmine and nefopam at the spinal dorsal horn.
