银杏叶提取物对白介素-1β损伤关节软骨细胞MMP-3和TIMP-1的影响 被引量:2

Effect of Ginkgo Biloba Extract (EGb761) on MMP-3 and TIMP-1 and Damaged Chondrocytes of Knee-joint Injured by IL-1β in Rabbits

摘要 目的:观察银杏叶提取物对白介素-1β损伤的关节软骨细胞的作用及对MMP-3、TIMP-1的影响。方法:在无菌环境下消化得到3个月龄雌性新西兰白兔膝关节软骨细胞,用第2代软骨细胞。软骨细胞与IL-1β(5μg/L)、IL-1β(5μg/L)加地塞米松(10-6mmol/L)、IL-1β(5μg/L)加银杏叶提取物(40mg、80mg、160mg/L)共同孵育,24h后ELISA测定受损伤软骨细胞培养液中的MMP-3及TIMP-1。结果:损伤后的软骨细胞培养液中的MMP-3增多,TIMP-1分泌较少。地塞米松同银杏叶提取物均可对抗IL-1β的效应,银杏叶提取物的这种效应有剂量依赖性。结论:银杏叶提取物通过调节受损伤后软骨细胞MMP-3和TIMP-1生成,保护IL-1β损伤的膝关节软骨细胞。 Objective To observe the protective effects of Ginkgo Biloba Extract (EGb761) on damaged chondrocytes of knee-joint injured by rratIL-1β in rabbits and to explore the protective mechanism of EGb761 through MMP-3 and TIMP-1. Methods Cartilaginous layers of knee were separated from three-month old Newzealand rabbit under semi-asepsis environment and then digested under sterile environment to get cartilaginous cells. The experimental cells were the second generation cartilaginous cells, identified with extracellular matrix in culture. These cells were observed by inverted microscopy and then were taken photos. Chondrocytes were divided randomly into six groups cultured with different cultures: rratIL-1β(5 μg/L) group, rratIL-1β(5 μg/L) with dexamethasone (10-6 mmol/L) group, rratIL-1β(5 μg/L) with EGb761(40 mg, 80 mg, 160 mg/L) group and control group. After 24 hours, rabbit matrix MMP-13 and TIMP-1 content were detected by ELISA assay. Results The production of MMP-13 was increased and TIMP-1 was decreased, Dexamethasone and EGb761 could rival the effect and EGb761 with dose-dependent response. Conclusion RGb761 could protect the damaged chondrocyte of articular genu injured by rratIL-1β in rabbit. The protective mechanism of EGb761 was to decrease production of MMP-13 and increase production of TIMP-1. EGb761 may be a new medicine to prevent osteoarthritis.
