摘要
取花魔芋(AmorphophalusKonjacK.Koch)长1.5cm的不定芽在附加1mg/L6-BA和0.05~0.1mg/LNAA的MS-NH+4培养基上,4℃黑暗保存180天后,存活达100%。不定芽在低温保存过程中,过氧化物酶活性升高,可溶性糖含量增加。培养基添加甘露醇后,过氧化物酶活性升高幅度变小,而可溶性糖含量进一步增加,保存后的花魔芋不定芽能快速恢复生长。甘露醇有利于芽的低温保存。
At 4℃ in darkness, Amorphophallus buds were able to be stored with 100% survial for 180 days on MS NH + 4 medium supplemented with 1 mg/L 6 BA, 0.05~0.1 mg/L NAA, and 3% (W/V) sucrose. Mannitol had been proved to be an appropriate growth retardant for low temperature storage of Amorphophallus buds. During the storage, the activity of POD and the content of soluble sugar in Amorphophallus buds had been increasing sharply. On MS NH + 4 medium with 10~15 g/L mannitol, the increase of the activity of POD in buds would slow down the increasing concentration of mannitol. On the contrary, the content of soluble sugar was accumulated with the presence of mannitol in medium. After storage for 90 and 180 days, buds grew quickly at the reculture period from 4th to 8th day, which would have relationship with the accumulation of soluble sugar in the buds during the storage.
