摘要 目的:观察部分丹参提取物对前脂肪细胞3T3-L1增殖、分化和分泌Leptin、PAI-1的影响,探讨其调脂作用的可能机制。方法:培养3T3-L1细胞,并用不同浓度的丹参提取物进行干预,以四甲基偶氮唑盐(MTT)法检测细胞增殖,用油红O染色和染色比色法分析脂肪细胞的分化程度。用ELISA法检测细胞培养上清中Leptin、PAI-1的含量。结果:(1)丹参素高浓度组明显抑制模型细胞增殖,中、低浓度组可显著抑制其分化;(2)丹参酮Ⅰ和丹参酮ⅡA明显抑制模型细胞的增殖,但对其分化无明显影响;(3)丹参素、丹参酮Ⅰ和丹参酮ⅡA均明显抑制脂肪细胞Leptin的分泌;(4)丹参素对脂肪细胞分泌PAI-1有明显抑制作用,丹参酮Ⅰ和丹参酮ⅡA作用不明显。结论:丹参提取物通过不同机制影响脂肪细胞,提示中药丹参可能通过多种机制作用于脂肪细胞而达到调脂作用。 Objective:To investigate the adjust fat possible mechanism of part Salvia extract through observing the effect on proliferation, differentiation and the excrete, of I_eptin,PAI - 1 of 3T3 - L1 preadipocyte. Methods: Culture 333 - L1 cells, using part Salvia extract of different dose to intervention, then through MTr method to detect the proliferation, oil red O staining method and spectrophotography to analyze the degree of differentiation, ELISA technique detect the Leptin, PAI - 1 contents in the mature fat ceils culture supernatant. Results : ( 1 ) In high - dose group of danshensu can inhibit the proliferation of 3T3 -L1 ,in middle and low- dose can inhibit the differentiation of 333 -L1 obviously; (2) Tan Ⅰ and Tan Ⅱ A can inhibit the proliferation of 3T3 -L1 obviously, but have no effect on the differentiation; (3)Dan-shensu,Tan Ⅰ and Tan Ⅱ A can all inhibit the excrete of mature fat cells of Leptin; (4) Danshensu can inhibit the excrete of mature fat cells of PAI- 1, but Tan Ⅰ and Tan Ⅱ A have no effect. Conclusion : The Salvia extract through different mechanism to effect preadipocyte, which suggest thin Chinese Salvia may through multiple mechanism to adjust fat.
