摘要
目的研究脑损伤后白细胞介素1β(interlukin-1,IL-1β)、丝裂原活化蛋白激酶p38(p38 mitogen-activa-ted protein kinases,p38 MAPK)及血管细胞间粘附分子-1(VCAM-1)在脑组织中的不同时相表达的特点及法医学意义。方法将75只清洁级SD大鼠随机分成正常对照组、手术对照组及损伤后0.5 h组、1 h组、3 h组、6 h组、12 h组、1 d组、3 d组、5 d组、7 d组、9 d组、14 d组。采用免疫组织化学染色技术检测脑组织中IL-1β、p38 MAPK及VCAM-1表达情况。结果对照组大鼠脑组织中IL-1β、p38 MAPK及VCAM-1均有少量表达。与对照组相比,损伤组脑组织中IL-1β的表达在损伤后0.5 h即升高,3 h达到高峰,6 h后逐渐降低,损伤后3 d降至正常水平;p38 MAPK的表达在损伤后1h开始升高,6h达到高峰,然后逐渐降低,损伤后5 d降至正常水平;VCAM-1的表达在损伤后0.5 h即升高,12 h达到高峰,1 d后逐渐降低,损伤后9 d降至正常水平。结论 IL-1β、p38 MAPK及VCAM-1在大鼠闭合性弥漫性脑损伤早期即迅速升高,后期逐渐恢复正常,3者在损伤后表达的时序性变化特点基本一致,共同参与脑损伤的病理生理过程;3者作为联合指标,在法医学实践中可判断损伤时间。
Objective To study the characteristics of expression and its forensic significance of interleukin-1 beta(IL-1β),mitogen-activated protein kinase p38(p38 MAPK) and vascular cell adhesion molecule-1(VCAM-1) in brain tissue with closed diffuse brain injury in different stage.Methods 75 Sprague SD rats were randomly divided into control groups(includes normal group and surgery group),and the injury groups(includes 0.5 h group,1 h group,3 h group,6 h group,12 h group,1 d group,3 d group,5 d group,7 d group,9 d group and 14 d group).Immunohistochemical staining detected the expression of IL-1β,p38 MAPK and VCAM-1 in brain tissue.Results In the control group,the expression of IL-1β,p38-MARK and VCAM-1 were little.Compared with the control group,the expression of IL-1β in brain tissue damage increased at 0.5 h,reached peak at 3 h,gradually decreased at 6 h and reached normal levels at 3 d;the expression of p38 MAPK began to increase at 1 h,reached peak at 6 h and then gradually decreased to normal levels at 5 d;the expression of VCAM-1 increased at 0.5 h,reached peak at 12 h,gradually decreased to normal at 9 d.Conclusion IL-1β,p38 MAPK and VCAM-1 in the closed diffuse brain injury in rats increased rapidly in early stage,gradually returned to normal in the late,the temporal changes of expression are consistent,which may participate in the pathophysiological process of brain injury;IL-1β,p38 MAPK and VCAM-1 as a co-indicators can be judged in the forensic injury time.
