摘要
目的从氧化应激以及炎症角度观察通脉养心丸对缺氧损伤的心肌细胞的影响,并探讨其可能的作用机制。方法采用大鼠心肌细胞原代培养技术并建立心肌细胞缺氧损伤模型,取成功培养的单层心肌细胞培养48h后分为空白对照组、缺氧损伤组、通脉养心丸组,通脉养心丸组分别给予通脉养心丸含药血清1g/kg、2g/kg、4g/kg、8g/kg,每组9孔。检测细胞上清液中超氧化物歧化酶(SOD)、谷胱甘肽酶(GSH)活性以及丙二醛(MDA)、白细胞介素6(IL-6)、白细胞介素1β(IL-1β)含量。结果与缺氧损伤组比较,通脉养心丸含药血清各剂量组能够显著升高SOD、GSH活性,降低MDA浓度(P<0.05或P<0.01);通脉养心丸含药血清4g/kg组能降低缺氧损伤引起的心肌细胞炎性因子IL-6、IL-1β浓度(P<0.05或P<0.01)。结论通脉养心丸具有抗心肌细胞缺氧损伤作用,抗氧化、抗炎可能是其作用机制之一。
Objective To observe the effect of Tongmai Yangxin Pills(Pills for removing obstruction of meridians and nourishing the heart)on cardiomyocytes with anoxic injury and study its mechanism on oxidative stress and inflammation.Methods A cardiomyocyte primary culture technique was applied to establish the anoxic injury models.After 48-hour culture,the successfully cultured monolayer myocardial cells were divided into control group,anoxic injury group,and Tongmai Yangxin Pills group.The Tongmai Yangyin Pill group were given medicine-contained serum of 1g/kg,2g/kg,4g/kg and 8g/kg,with 9 samples in each group.Subsequently,SOD and GSH activity and MDA content were measured,and IL-6 and IL-1β concentration was measured.Results Compared with the anoxic injury group,all Tongmai Yangxin Pills groups could increase the SOD and GSH activity,and decrease the MDA content(P0.01 or P0.05).The Tongmai Yangxin Pills 4g/kg group could decrease the cardiomyocyte inflammatory factors IL-6 and IL-1β concentration due to anoxic injury(P0.05 or P0.01).Conclusion Tongmai Yangxin Pills functions to protect the cardiomyocytes from hypoxia injury.One of its mechanisms is anti-oxidation and anti-inflammation.
