摘要
以啶虫脒原药与β-巯基丙酸为起始原料,在热碱作用下合成啶虫脒的人工半抗原.再采用EDC两步法使该半抗原与牛血清蛋白偶联得到啶虫脒的完全抗原.通过质谱、核磁共振谱、红外光谱和紫外光谱对啶虫脒人工半抗原进行结构表征,利用紫外光谱对合成的半抗原与载体蛋白的偶联情况分析,证明啶虫脒人工半抗原合成成功、啶虫脒人工半抗原与载体蛋白偶联成功,偶联比为1∶18.5.通过啶虫脒免疫抗原免疫实验小鼠,获得到了高效价抗血清,最终证明啶虫脒半抗原、人工抗原合成成功.
Acetamiprid's artificial hapten has been synthesized by using acetamiprid reacted with β-mercaptopropionic acid in the hot alkalic solution.And the hapten was conjugated with bovine serum albumin to form the artificial complete antigen.And then the molecular and characteristic ion peak of the artificial hapten was identified with MS(ESI).Expectation of the hapten structure was indicated by MS,NMR,IR and UV.And that the hapten with bovine serum albumin was coupled successfully was also showed in their IR and UV.And the coupling ratio was 1:18.5.and above all was proved by the animal immune experiment.
