摘要
近年来,在DNA检测中荧光光谱法被广泛应用.新型荧光探针化合物的发现成为当前研究的热点,本文以自主发现的荧光分子4H,4'H-2,2'-双色烯-4,4'二酮(简称BCDO)为荧光探针,采用荧光猝灭法,通过体系荧光光谱的测定,对其与小牛胸腺DNA的作用方式进行了研究.通过DNA溶液浓度对荧光分子BCDO的影响实验,F0/F~C曲线的绘制,发现BCDO与DNA的相互作用为静态猝灭过程,其猝灭常数Kq在106~108数量级之间,大于106数量级,说明两者之间有很强的亲和能力.由Na+对BCDO-DNA体系的影响实验进一步说明了BCDO与DNA之间的相互作用方式为静电作用.
In recent years,fluorescence spectrometry was widely used in the test of DNA.The discovery of new fluorescent compounds was currently a hot research.In this paper,self-made 4H,4′H-2,2′-bichromene-4,4′-dione(referred to BCDO) was used as a fluorescent probe,the interaction of BDCO and DNA was investigated by fluorescence spectrometry.Through the experiment on the fluorescence quenching spectra of BCDO by DNA at different concentrations,drawing F0/F~C curve,it was found that the interaction of BCDO and DNA is a static quenching process,the quenching constant was in excess of 106,it was showed that BDCO with DNA have strong affinity .The Na+ experimental results further indicated that BDCO interacted with DNA in electro static binding mode.
