GRIM-19及其靶基因产物STAT3与肿瘤关系的研究进展

摘要 干扰素/维甲酸联合应用诱导细胞凋亡相关的基因19(gene associated with retinoid-interferon-induced mortality-19,GRIM-19)是一种新的抑癌基因,通过多种机制参与细胞凋亡,是信号转导及转录活化因子3(signal transducers and activators of transcription 3,STAT3)的特异性抑制蛋白,在多种肿瘤组织中低表达。STAT3及其介导的信号转导通路参与调节细胞的增殖、凋亡与分化,介导细胞的恶性转化,并与肿瘤的发生发展关系密切。恢复GRIM-19的表达通过抑制STAT3的表达和激活而显著抑制肿瘤细胞的生长。 GRIM-19(gene associated with retinoid-interferon-induced mortality-19)gene is a new tumor-suppressor gene,involved in apoptosis through multiple mechanisms.It is a specific protein to inhibit signal transducers and activators of transcription 3(STAT3) and low expression in a variety of tumor tissues.STAT3 and its mediatedsignal transduction pathway involved in regulating cell proliferation,apoptosis and differentiation,cell-mediated malignant transformation,and is closely related with the tumor development.The expression of GRIM-19 restored by inhibiting the expression and activation of STAT3 significantly inhibited tumor cell growth.
