摘要
根据GenBank中猪繁殖与呼吸综合征病毒(PRRSV)VR-2332毒株基因序列设计合成了6对引物,应用RT-PCR方法对PRRSVGS株部分片段进行基因的扩增、克隆和测序,将测序结果应用Blast和DNAStar软件进行拼接,并与国内外分离毒株进行核苷酸以及推导氨基酸序列同源性比较.结果发现PRRSVGS毒株结构蛋白基因全长3819bp,包括ORF2、ORF3、ORF4、ORF5、ORF6和ORF7共6个阅读框,分别编码GP2、GP3、GP4、GP5、M和N蛋白;与BJ-4、CH-1a、HB-2(sh)/2002和VR-2332美洲株等核苷酸的同源性在87.9%~97.1%,推导的氨基酸同源性在88.3%~97.1%;而与LV和Euro毒株的同源性差异显著,核苷酸同源性在56.0%~70.0%,氨基酸同源性在54.9%~77.6%.构建的系统发育树证明该毒株在基因型上属于美洲型.
According to the sequence of porcine reproductive and respiratory syndrome virus (PRRSV) VR-2332 strain published in Genbank,six pairs of primers were designed to amplify the structural protein genes of PRRSV GS strain by RT-PCR.The sequences were compared with the sequences of other PRRSV strains in GenBank by Blast and DNAStar software.The results showed that the complete structural protein genes of PRRSV GS strain was 3 819 bp,the open-reading frame include ORF2,ORF3,ORF4,ORF5,ORF6 and ORF7,coding in GP2,GP3,GP4,GP5,M and N.The ORF2~ORF7 of the PRRSV GS strain shared 87.9 %~97.1 % of nucleotide homology and 88.3 %~97.1 % of amino acid homology with that of North American genotype (BJ-4,CH-1a,HB-2sh/2002 and VR-2332),respectively.And they shared only 56.0 %~70.0 % nucleotide homology and 54.9 %~77.6 % of amino acid homology with that of LV and Euro strain.The phylogenetic tree analysis revealed that PRRSV GS strain was clustered within North American genotype.
