摘要 目的 探讨硫酸乙酰肝素蛋白聚糖(HSPG syndecan-1)表达水平与肺癌分型的相关性,以及HSPG syndecan-1 蛋白和增殖诱导配体(APRIL)蛋白在肺癌细胞中的相互作用.方法 回顾性分析上海市普陀区人民医院67例肺癌组织标本,实时荧光定量PCR,Western blot和流式细胞术检测HSPG syndecan-1在肺癌组织的表达情况,激光共聚焦技术检测HSPG syndecan-1和APRIL的相互作用情况.结果 HSPG syndecan-1基因的mRNA、蛋白在肺癌中较正常肺组织高表达;在各种类型的肺癌组织中,HSPG Syndecan-1基因在高分化肺癌的表达高于低分化肺癌.HSPG Syndecan-1蛋白和APRIL蛋白在肺癌细胞中存在相互作用.结论 HSPG Syndecan-1蛋白在肺癌中的表达和肺癌细胞的分化程度相关,与APRIL蛋白存在相互作用. Objective To investigate the relationship between the level of syndecan-1 expression and types of lung cancer and to investigate whether syndecan-1 protein and APRIL protein interacts in lung cancer cells. Methods A total of 67 samples of lung cancer were analysed retrospectively to determine the expression of syndecan-1 in various types of lung cancer cells by fluorescence quantitative RT-PCR,Western blot and flow cytometry. Immunofluorescence double labeling reaction were performed to determine whether syndecan-1 protein and APRIL protein could interact, Results Compared to normal lung tissue,Syndecan-1 mRNA and protein was highly expressed in lung cancer cells. In various types of lung cancer cells ,Syndeean-1 gene expression level was higher in well differentiated lung cancer cells than in poorly differentiated cells. There was possibility of interaction between Syndecan-1 protein and APRIL protein in lung cancer cells. Conclusion The expression of Syndecan-1 in lung cancer is related to the degree of differentiation of lung cancer cells. There is interaction between proteins of Syndecan-1 and APRIL.
