胡萝卜对苯乙酮及其衍生物的不对称催化还原反应研究 被引量:3

摘要 [目的]利用廉价的胡萝卜根部细胞组织进行苯乙酮及其衍生物的生物催化还原反应,制备S-(-)-1-苯乙醇及相关产物。[方法]考察反应时间、底物浓度、反应温度以及摇床转速对该不对称催化还原反应的影响,并利用红外光谱仪、核磁共振仪等对所得产物结构进行鉴定。[结果]在环境温度25℃,70 ml水中加入0.05 g苯乙酮,摇床转速为150 rpm,反应72 h的条件下,S-(-)-1-苯乙醇收率最高,达71%,ee值为83.9%。在该条件下,对苯乙酮衍生物进行催化还原研究,最终选择性地还原得到S构型的1-苯乙醇衍生物,收率和ee值依次在60%和90%左右。[结论]整个反应体系条件温和,成本廉价,具有较高的实用前景。 [Objective]Taking the low-priced carrot root cells as catalyst,the biocatalytic reduction of acetophenone and acetophenone derivatives were carried out,S-(-)-1-phenylethanol and relevant products were prepared.[Method] The effects of reaction time,substrate concentration,reaction temperature and shaking speed on the reaction were investigated,and the structure of reaction products were identified by IR,1HNMR and so on.[Result]The results showed that the conversion could reach the maximum yield of 71% and ee value of 83.9% at 25 ℃,0.05 g acetophenone added to 70 ml water,shaking speed of 150 rpm,reaction time of 72 hours.Under the best reaction conditions,the catalytic reduction of acetophenone derivatives were studied.Finally,the S-1-phenylethanol derivatives were prepared(yield about 60%,ee value about 90%).[Conclusion]The whole reaction system had a promising practical future with its mild reaction conditions and lower price.
