摘要
以戊二醛交联的牛血清白蛋白和β-环糊精作为固定化载体固定醇氧化酶和甲醛脱氢酶,利用二茂铁作为电子媒介体,研制出在乙醇存在的条件下,测定甲醇浓度的生物传感器,同时探讨了工作电位、pH、干扰物质等对该生物传感器的影响。结果表明,在工作电位为0.35 V;pH 7.8时,传感器有良好的响应性能,响应电流与甲醇浓度在0.005%~0.08%时呈线性关系,最低甲醇检测浓度在0.002%,达到95%稳态的时间不超过20 s,且重现性良好,具有较好的应用前景。
Alcohol oxidase,together with formaldehyde dehydrogenase,was cross-linked with bovine serum albumin(BSA).Then it was immobilized by glutaraldehyde and β-cyclodextrin as matrix.The ferrocene was an electron transfer mediator.The methanol biosensor was constructed to determine the content of methanol in ethanol.Meanwhile,the effects of operating potential,pH,interfering substances on the biosensor were discussed.The biosensor showed good performance when operating potential was at 0.35V and pH value was 7.8.And the response current had a good linear relationship with the concentration of methanol between 0.005 %~0.08 %.The lowest detection concentration of methanol was 0.002 % and the detection time was less than 20 s as it reached 95 % steady state.The biosensor had good reproducibility and good application foreground.
