摘要
目的:观察电刺激大鼠黑质网状部(substantia nigra pars reticulata,SNr)对脚桥核(pedunculopontine nu-cleus,PPN)神经元自发放电活动的影响,进一步探讨脑内电刺激治疗帕金森病(Parkinson’s disease,PD)的机制。方法:应用细胞外记录方法观察不同频率电刺激大鼠SNr对PPN神经元自发放电的影响。结果:实验记录了大鼠46个PPN神经元的自发放电,其放电频率在2.4~55.4 Hz之间,平均为(21.3±13.8)Hz;低频电刺激(5~10Hz)大鼠SNr时,PPN神经元的自发放电无明显变化(P〉0.05),随着刺激频率(20~200 Hz)的增加,大多数PPN神经元的自发放电活动受抑制,神经元放电频率较刺激前明显降低,且在100 Hz时的抑制作用最强(P〈0.05)。结论:高频电刺激大鼠SNr对PPN神经元自发放电的影响主要为抑制,提示高频电刺激SNr可通过抑制PPN神经元活动参与PD的治疗。
Objective:To study the effect of electrical stimulation of the substantia nigra pars reticulate(SNr) on the neuronal activities of pedunculopontine nucleus(PPN)in rats,furthering the mechanism of deep brain stimulation in the treatment of Parkinson's disease.Methods:Electrophysiological recordings were made in anaesthetized rats to investigate the effects of SNr electrical stimulation with different frequencies on the firing rate of the PPN neurons.Results:For SNr stimulation at low frequency(5-10 Hz),there was no effect on the spontaneous firing activities of PPN neurons(P0.05).With the increase of stimulation frequency(20-200 Hz),the majority of neurons recorded in the PPN were inhibited.Conclusion:High frequency stimulation of the SNr induced an obvious decrease in activity of most neurons recorded in the PPN.The present data provide foundation for future investigations afferent pathways of SNr-PPN in the treatment of Parkinson's disease.
