摘要
为降低口蹄疫O型和Asia 1型二价灭活疫苗使用过程中存在的免疫副反应,本研究采用超滤技术对抗原进行浓缩纯化处理,制备浓缩抗原疫苗,通过测定制苗用病毒液LD50值和浓缩抗原疫苗内毒素含量,进行牛副反应试验,并与未浓缩的普通疫苗进行比较。结果表明,制苗用Asia 1/JSL06株和OS/99株病毒液经浓缩纯化处理后LD50值比常规处理LD50分别提高了1.0和1.25个滴度,内毒素值显著低于普通灭活苗内毒素值(p<0.01),接种本动物后未出现免疫副反应。本研究结果表明,使用超滤技术浓缩纯化抗原可有效降低口蹄疫灭活苗免疫副反应。
In order to reduce immunological side effects of bivalent vaccine against type O and Asia-1 foot and mouth disease, a concentrated antigen vaccine was prepared using ultrafiltration technology for enrichment and purification of antigen. The LD^o values of the virus solution for vaccine preparation and the endotoxin concentration of the concentrated antigen vaccine were measured, The in vivo side effects of the vaccine in bovine were compared with those of normal un-concentrated vaccine. The results showed that, after concentration and purification, the LD50 values of Asia-1/JSL06 strain and OS/99 strain increased by 1.0 and 1.25 on a log scale, respectively. The endotoxin was significantly reduced compared with that of inactivated vaccine. No obvious immunological side effects were detected in animal test. It was concluded that the ultrafiltration technology could be successfully applied to concentrate and purify FMD antigen to produce effective and safe inactivated FMD vaccine with undetectable immunological side effects.
